چکیده

It has been shown that changes in fatty acid turnover are an early event in the response in vitro of animal lymphocytes to mitogens (Resch & Ferber, 1972). Fatty acids added to the medium in ethanol inhibit the increase in uridine and thymidine uptake in human lymphocytes, which is a normal part of the response (Meriin ef al., 1974). We have previously reported that even when bound to albumin, single fatty acids still inhibit the blast transformation of human lymphocytes (Weyman et al., 1975), but that certain combinations of acids inhibit little or not at all, whereas with other combinations the inhibition is greater than with single acids. In the human, mixtures containing only saturated acids and those containing only unsaturated acids gave considerable inhibition, but mixtures in which a balance was maintained between saturated and unsaturated acids inhibited comparatively little. An exception was found to this rule, in that myristate did not itself inhibit, but relieved the inhibition by both saturated and unsaturated acids. We have now extended this work to examine the response of lymphocytes of several species to the inclusion of acids in the medium. The results are summarized in Table 1 (the results from the human are included for comparison).

Glucose Metabolism of Normal Human Lymphocytes

1. Phytohaemagglutinin induced early changes in the catabolism of glucose by normal human lymphocytes suspended in a bicarbonate buffer. During 4hr. incubation glucose utilization was almost doubled. 2. The rates of several reactions in the metabolism of glucose were estimated. Total pyruvate formation, lactate production and fatty acid synthesis were stimulated to the same degree as was glucos...

Fatty acid inhibition of lipopolysaccharide-stimulated B lymphocyte proliferation.

We have recently shown that saturated, monounsaturated and polyunsaturated fatty acids, including those of fish-oil origin, inhibit [ 'Hlthymidine incorporation into DNA of concanavalin A (Con A)-stimulated rat lymph node lymphocytes [ 1, 21. All nine fatty acids tested inhibited the response to mitogen, but the extent of inhibition was dependent upon the fatty acid concentration used, the time...

Lipid Mixtures Containing a Very High Proportion of Saturated Fatty Acids Only Modestly Impair Insulin Signaling in Cultured Muscle Cells

In vitro examinations of the effect of saturated fatty acids on skeletal muscle insulin action often use only one or two different fatty acid species, which does not resemble the human plasma fatty acid profile. We compared graded concentrations (0.1-0.8 mM) of 3 different lipid mixtures: 1) a physiologic fatty acid mixture (NORM; 40% saturated fatty acids), 2) a physiologic mixture high in sat...

Determination and study the fatty acid contents and their seasonal variations by temperature of a dominant bivalve(Callista umbonella) of Haleh Creek

In this study fatty acid contents, because of the importance in human’s life, and their seasonal variations of a dominant bivalve of Haleh Creek were determined for the first time. Fatty acid identification was done by GC-MS (Gas Chromatography–Mass Spectrometry) method. After collecting and dissecting the species from the shells, samples weighed and frozen for further experiments. All the samp...

In vitro folate deficiency induces apoptosis by a p53, Fas (Apo-1, CD95) independent, bcl-2 related mechanism in phytohaemagglutinin-stimulated human peripheral blood lymphocytes.

In vitro folate deficiency is associated with S phase accumulation and apoptosis in various cell types. To investigate the role of p53 and two apoptosis-related molecules, bcl-2 and Fas antigen (Apo-1, CD95), in the mechanism whereby folate-deficient lymphocytes accumulate and undergo apoptosis in the S phase, normal human peripheral blood lymphocytes were cultured for 3-9 d in control medium o...
